I have sent several requests asking how to cancel my booking. I have never received a response. My room is not available – please remove it from booking.com immediately.

You can't just cancel a confirmed booking. You'll need to contact Booking.com that you got overbooked and that they should find another accommodation for the guest (if you can not). You will have to pay the commission for this booking and possible some extra fees.
You can request to cancel in extranet and contact the guest to accept it. If he/she won't accept then Booking.com will have to step in.
