From the moment you have put availabilities on your Booking.com calendar, the room is considered as available. If a reservation is done, it has to be honored or see what you can do with our customer service.

Imho the web backend of booking.com is WAY too complicated and SHOULD include a dashboard wide settings search (you know you search for "instant" and it should filter a list of possible options the user can set and when clicking on an list item directly go to url and highlight where this setting can be found... like any Android is doing that)

To modify your settings and en/disable "instant booking", follow the below steps:
1 - Log in your Partner account (https://admin.booking.com/hotel/hoteladmin/extranet_ng/manage/home.html…) and go to:
Property > Policies > How you receive bookings
2- Select "Edit"
3- Select between:
"All guests can book instantly"
OR
"All guests will need to request to book"
4- Select "Save"
